In this talk, I will provide an updated overview of the neurobiological actions of the growth hormone secretagogue receptor (GHSR), with a particular focus on its endogenous ligands—ghrelin and Liver-Expressed Antimicrobial Peptide 2 (LEAP2)—as well as its putative ligand-independent actions. I will discuss recent advances in our understanding of how GHSR signaling influences brain circuits involved in the regulation of food intake, food reward, and stress. Special emphasis will be placed on novel findings from our group, including insights into the accessibility of circulating ligands to centrally expressed receptors, the neuronal mechanisms underlying food intake and stress responses, and the dynamic regulation of LEAP2. These findings expand our perspective on GHSR function and open new avenues for exploring its physiological and therapeutic relevance.
